Unlock the listed character by completing the corresponding task.
- BabyDaisy - Using Wario's magnet, pull Daisy’s baby rattle from the fountain at Peach Garden. Then, have Yoshi go through the manhole coveron the left side of the garden to find a crying baby. Give Baby Daisy the rattle.
- Baby DK - At DK Jungle, use Yoshi's power to free Baby DK from the palm tree to unlock Baby DK. Alternatively, defeat DK's team.
- Baby Luigi - In Mario's stadium, pull the levers to connect the bridge.
- Baby Mario - Unlock Baby Luigi, then save Baby Mario at the field where you battle Bowser Jr.'s team.
- Baby Peach - Use Princess Peach's heart power on the grass Toad statue at Peach Garden.
- Birdo - Defeat Bowser Jr. at Yoshi Park.
- Blooper- Have Princess Peach use her heart power on the bubbles in the water at the dock in Mario Stadium. Alternatively, complete all tutorials in Practice mode.
- Blue Noki - At Mario's stadium, complete Blue Noki's task.
- Blue Pianta - At Mario's stadium, get the key and save Blue Pianta from the shop.
- Blue Shy Guy - At Yoshi's ballpark, shoot the Piranha Plants.
- Boo - At Wario city, solve the block puzzle with DK and Wario.
- Daisy - Complete Peach's Stadium.
- Funky Kong - Save Funky Kong from the Hammer Brother at DK Jungle.
- Goomba - Help Goomba trace his shop at Wario City.
- Gray Shy Guy - At Yoshi Park, have Peach get Gray Shy Guy out of the log.
- Green Koopa - Free Green Koopa from the poster at Wario City.
- Green Noki - Unlock and have Blue Noki on your team, then talk to Green Noki at the beach at Mario's Stadium.
- Green Shy Guy - Have Peach get Green Shy Guy out of the log at Yoshi Park.
- Green Toad - Complete To adette's puzzle, then search the bush at Peach's Stadium.
- Green Yoshi - Free Green Yoshi at the manhole at Yoshi's ballpark.
- King Boo - Talk to the Tiny Boos in a chest at Wario City.
- King K. Rool - Defeat all four Kritters at DK Jungle. Then, go to the waterfall, and defeat King K. Rool.
- Kritter - Defeat King K. Rool at DK Jungle.
- Light Blue Yoshi - Rescue Light Blue Yoshi from Green Magikoopa in Yoshi Park.
- Monty Mole - Use Peach on the Monty outline on the beach at Mario Stadium.
- Paragoomba - Use Yoshi's ground pound on one of the palm trees in Wario City.
- Paratroopa- Defeat the minion guarding the lighthouse at Wario City.
- Peach - Complete Peach's Stadium.
- Pink Pianta - At Mario's Stadium find Pink Pinata in a palm tree on the beach.
- RedNoki - Unlock and have both Blue Noki and Green Noki on your team.Then, talk to Red Noki on the giant clam at Mario's Stadium.
- Red Paratroopa - At Yoshi park, find the paintbrush and use it to paint the giant billboard at Wario City.
- Red Shy Guy - Shoot the Piranha Plants at Yoshi's Ballpark.
- Red Yoshi - Find the brush that Red Yoshi lost, then find him at Yoshi's Stadium.
- Tiny Kong - Defeat Bowser Jr. at DK Jungle.
- Toad - Start Peach's Stadium.
- Toadette - Talk to Toadette at Peach's Stadium.
- Waluigi - Find Waluigi on top of a skyscraper in Wario City.
- Wiggler - At Yoshi park, reset the roller coaster that Wiggler is riding. Alternatively, win a game at Toy Field.
- Yellow Pianta - Go to Mario Stadium to find Yellow Pinata in a barrel.
- Yellow Yoshi - At Yoshi park, use Yoshi to get Yellow Yoshi out of a tree.
Secret Shop items:
At the secret shop, unlock the listed item by purchasing the corresponding item(s). - Star Candy - Make any purchase at the Secret Shop.
- Star Candy (x2) - Purchase a Star Candy.
- Super Star - Purchase three Star Candy (x2).
Error items:
Unlock the listed error item by using Wario's treasure opening ability at the corresponding location.

Win at Bowser Jr.'s Playroom in challenge mode to unlock night mode at all stadiums except Bowser's stadium and Luigi's mansion.

- Blooper Baserun (Day) - Unlock Daisy’s Cruiser.
- Bomb-omb Derby (Night) - Defeat Bowser Jr. in Challenge Mode.
- Castle Pinball (Night) - Unlock Bowser’s Castle.

To become a member of the Secret Shop, open up the chest in DK’s Stadium during Challenge Mode with Wario.
